Environmental stimuli and intestinal stem cell behavior

چکیده
The intestine carries out important functions related to digestion and absorption. It is composed of three distinct layers, an outer muscle layer, a mesenchymal layer and the epithelial layer. The epithelial layer forms the protective barrier that faces the luminal content of the intestine. In order to maintain barrier function the epithelial layer needs constant replenishment. This is ensured by continuous cellular replication in proliferative crypt compartments. Following exit from the crypt, cells adopt fates along either secretory or absorptive lineage and will, after three to four days, be exfoliated into the lumen of the intestine from the tips of the villi. Intestinal stem cells located at the bottom of the proliferative crypt compartment ensure lifelong maintenance of the organ (Fig. 1A). Adult stem cell niches are far more heterogeneous than previously anticipated. The intestinal stem cell niche can be subdivided by the relative position within the crypt. Stem cells located in position +4, just above secretory Paneth cells, express HopX, Bmi1 and Tert. These cells are generally less mitotically active than Lgr5-expressing stem cells located at the bottom of the proliferative crypts intercalated between Paneth cells (Fig. 1A). It has been argued that both populations represent the most primitive stem cell; however, recent studies suggest that stem cells can interconvert between the two states (Fig. 1B). Heterogeneity of the Level of Activity of Lgr5+ Intestinal Stem Cells
Intestinal stem cells (ISCs) are a group of rare cells located in the intestinal crypts which are responsible for the maintenance of the intestinal homeostasis and intestinal regeneration following injury or inflammation. Lineage tracing experiments in mice have proven that ISCs can repopulate the entire intestinal crypt. It is noteworthy that in such experiments, only a subset of intestinal cr...

Dental Pulp Stem Cells (DPSCs) can be found within the cell rich zone of dental pulp. These stem cells, under specific stimuli, differentiate into many cell types which have wide therapeutic applications. The dental stem cells are derived from both deciduous and permanent teeth. The viable dental stem cells are very simple to collect, without any mortality and morbidity. Dental pulp stem c...

Cancer stem cells (CSCs) are unique subpopulations that have the capacity to drive malignant progression with renewal abilities. Recently the role of some of CSCs in gastric adenocarcinoma has been studied. This study was performed in order to evaluate CD44 and CD133 expressions by immunohistochemistry in 95 primary gastric adenocarcinoma and their relation to clinical and pathological paramete...
متن کاملIdentification of diagnostic biomarkers by bioinformatics analysis in the inflamed and non-inflamed intestinal mucosa in Crohn\'s disease patients
Background: Crohn's disease (CD) is a type of inflammatory bowel disease (IBD) which despite the unknown details is generally related to genetic, immune system, and environmental factors. In this study, we identify transcriptional signatures in patients with CD and then explain the potential molecular mechanisms in inflamed and non-inflamed intestinal mucosa in these patients. Materials and Me...
